- Synthetic ORF encoding the BPV1 L1 protein was constructed with Invitrogen custom oligos purified with PAGE. Vector pUb6-V5-HisA was used to construct the target plasmids to express the ORF with Ecdysone inducible expression system. CV-1 and COS-7 cells were transfected using Lipofectamine Plus. 293 cells were transfected using Lipofectamine 2000.

- Sf9 cells were grown in Grace's medium, D-MEM contained 5% FCS and pen-strep. The 6-5 cell line was maintained with G418. Restriction endonucleases, other DNA-modifying enzymes and DNA primers were purchased from Invitrogen. For preparation of a recombinant baculovirus expressing UL12.5, the Bac-to-Bac system was used. The cells were transfected with a total of 1 µg of plasmid DNA using Lipofectamine PLUS.
